چکیده

common to both these compounds (Heath & Wildy, 1956a, b). These results indicated that this fungus forms ergothioneine by methylation and sulphuration of histidine. Subsequent work (Wildy & Heath, 1956, 1957) revealed that [2(ring)-14C]histamine would not serve as a precursor of the imidazole nucleus of either histidine or ergothioneine. Various metabolites of histamine, imidazoles possessing modified side chains, also failed to provide the requirements of an ergothioneine precursor. In order to confirm that histidine was a direct precursor of ergothioneine, experiments were carried out with histidine labelled both in the side chain and the imidazole ring. A preliminary report of this work has already been published (Heath & Wildy, 1957).

Biosynthesis of ergotamine by Claviceps purpurea (Fr.) Tul.

High-yielding strains of Claviceps purpurea (Fr.) Tul, grown on a defined medium, have been used for a study of the biosynthesis of the peptide ergot alkaloid, ergotamine. l-[U-(14)C]tryptophan, dl-[2-(14)C]mevalonic acid lactone, sodium [2-(14)C]acetate, sodium [(14)C]formate and the methyl group of l-[methyl-(14)C]methionine were efficiently incorporated into the peptide alkaloids and specifi...

Crystallization and preliminary X-ray analysis of the ergothioneine-biosynthetic methyltransferase EgtD.

Ergothioneine is an amino-acid betaine derivative of histidine that was discovered more than one century ago. Despite significant research pointing to a function in oxidative stress defence, the exact mechanisms of action of ergothioneine remain elusive. Although both humans and bacterial pathogens such as Mycobacterium tuberculosis seem to depend on ergothioneine, humans are devoid of the corr...

Improved synthesis of the super antioxidant, ergothioneine, and its biosynthetic pathway intermediates.

Ergothioneine and mycothiol are low molecular mass redox protective thiols present in actinomycetes, in particular mycobacteria. We report the improved chemical synthesis of ergothioneine (ESH) and biosynthetic pathway intermediates using either histidine or ESH as the starting material. The detailed mechanism of ESH biosynthesis has not yet been completely elucidated and substrates for enzymes...

The reaction sequence in ergothioneine biosynthesis: hercynine as an intermediate.

Ergothioneine can be synthesized by the mold, Neurospora crassa, from the amino acids, histidine, methionine, and cysteine (1). The intact histidine molecule is used in the synthesis, and the three methyl groups of the betaine moiety can be derived from methionine by a triple transmethylation process (2). Cysteine appears to be an immediate precursor of the sulfur atom of the thiolimidazole rin...
